ab28049 staining mouse intestinal tissue sections by IHC-P. Sections were PFA fixed and subjected to heat mediated antigen retrieval by microwaving in boiling water for 15 minutes prior to permeabilization for 15 min in 0.2% PBST and blocking with 20% goat serum for 30 minutes at RT. The primary antibody was diluted 1/400 and incubated with the sample for 15 hours at 4°C. An Alexa Fluor® 488 conjugated goat anti-rabbit antibody was used as the secondary. Specific staining can be seen in the perinuclear region of the enterocytes (red). Nuclei were counterstained with PI (blue).See Abreview
ab28049 staining mouse cortical oligodendrocytes from vibratome sections by IHC-Fr. Sections were fixed in Somogyi (PFA + glutaraldehyde + picric acid) or PLP (Periodate-lysine-PFA) and permeabilized in TX-100, prior to blocking with 2.5% serum for 2 hours at 25°C. The primary antibody was diluted 1/1000 and incubated with the sample for 18 hours. A TRITC conjugated goat anti-rabbit IgG antibody was used as the secondary.See Abreview
