IHC image of ab26127 staining Histone H3 in Human normal colon formalin fixed paraffin embedded tissue* sections, performed on a Leica Bond. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab26127, 7µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. ab26127 staining Histone H3 (phospho T45) in HEK 293T cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilized with 1% Triton X-100 and blocked with 3% BSA for 1 hour. Samples were incubated with primary antibody (1/50 in PBS + 3% BSA) for 1 hour. ChIP analysis using ab26127 binding Histone H3 in Human RWP1 whole cell lysate. Cells were cross-linked for 5 minutes with 4% PFA.
