Immuofluorescent staining for Nicotinic Acetylcholine Receptor alpha 7 in the rat cerebellum using the Rabbit polyclonal to Nicotinic Acetylcholine Receptor alpha 7 (ab23832; Image is taken with a X40 objective). Nicotinic Acetylcholine Receptor alpha 7 staining has been previously described in Purkinje cells (arrows) and in the processes of these cells. Pre-incubation of ab23832 with the immunizing peptide (ab24285) quenches the immunostaining (data not shown). ab23832 was used at 1/300 (1 µg/ml) incubated overnight at room temperature. Secondary antibody used was anti-rabbit Alexa Fluor 488 at 1/1000 incubated for 2 hours at room temperature. Rat brain tissue was perfusion fixed with 4% PFA followed by overnight post-fixation in the same fixative, cryoprotected in 20% sucrose and frozen in OCT. 30µm coronal sections were cut on a cyrostat and immunohistochemistry performed by the 'free floating' technique.
