ab50008 (1:500, 1hr/ 37oC) staining Nuclear pore complex proteins in HeLa cells (green). Cells were fixed in PHEM buffer (45 mM Hepes pH 6.9, 45 mM Pipes pH 6.9, 5 mM MgCl2, 10 mM EGTA, 4% PFA) /0,2% Triton x-100, 10 min at room T°, then quickly wash 3 x in PBS. ) Good results are also obtained after straight PFA fix: fix in 3.7% PFA / 30 mM sucrose for 10 min at room T°, quickly wash 3 x in PBS, then incubatein PBS/ 0.1% TritonX-100 for 5 min and further in 0.1 M glycine for 10 min.Image kindly provided by R.
