ab79389 at 1/1000 dilution staining Pax2 in human fetal kidney (image A), human normal kidney (image B) and human renal cell carcinoma (image C) by Immunohistochemistry, Paraffin-embedded tissues.

Overlay histogram showing HEK293 cells stained with ab79389 (red line). The cells were fixed with methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab79389, 1/50 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit monoclonal IgG (1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a significantly decreased signal in HEK293 cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.

Immunohistochemical analysis of Human malignant melanoma tissue, staining Pax2 with ab79389. Antigen retrieval was performed via heat mediation in a citrate buffer (pH 6). Sections were blocked with 0.1% Triton X-100/PBS containing 1% BSA and 10% horse serum for 1 hour. Samples were incubated with primary antibody overnight at 4°C. A Cy3®-conjugated goat anti-rabbit IgG was used as the secondary antibody.
