ab49507 staining Tartate Resistance Acid Phosphatase in Human osteoclast cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 3% BSA for 2 hours at 4°C. Samples were incubated with primary antibody (1/100 in 0.1% Triton + 3% BSA) for 12 hours at 4°C. An undiluted Alexa Fluor® 555-conjugated Donkey anti-mouse IgG polyclonal was used as the secondary antibody.See Abreview

Immunohistochemical staining (Paraffin section) for Tartrate Resistant Acid Phosphatase using ab49507 with Human osteoclastoma as positive control. Note intense cytoplasmic staining of osteoclasts.

ICC/IF image of ab49507 stained MCF7 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab49507, 1/1000 dilution) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
