NRK cells were stained with anti-TGN46 ab2809 (red) anti-beta-COP ab2899 (green) and DAPI.
Immunofluorescent analysis of Phalloidin (green) and TGN46(purple) in U2OS cells. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in PBS for 10 minutes at room temperature and blocked with 2% BSA in PBS + 0.1% Triton X-100 for 30 minutes at room temperature. Cells were probed with a TGN46 monoclonal antibody (ab2809) at a dilution of 1:75 for at least 1 hour at room temperature and incubated with DyLight 680 goat anti-mouse IgG secondary antibody at a dilution of 1:250 for 30 minutes at room temperature. Actin was stained with DyLight 488 Phalloidin at a dilution of 1:300 (1 unit/ml final concentration) for 30 minutes. Images were taken at 20X magnification.
Immunofluorescent analysis of TGN46 using anti-TGN46 monoclonal antibody (ab2809) shows staining in NS-1 Cells.
HeLa and COS7 cells were rinsed with PBS and fixed with -20�C methanol for 5 minutes. Cells were incubated with anti-TGN46 antibody (ab2809) diluted 1:100 in PBS containing 0.2% BSA for 1 hour. The cells were then rinsed in PBS and incubated with goat anti-mouse IgG conjugated to rhodamine for 30 minutes in PBS containing 0.2% BSA.
ICC/IF image of ab2809 stained rat PC12 cells. The cells were 4% PFA and 100% Methanol fixed (10 min) and then incubated in 1% BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab2809, 5 µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue).
ab2809 staining Chinese Hamster Ovary cell by Immunocytochemistry/Immunofluorescence. The cells were paraformaldehyde fixed and permeabilized in 0.5% Triton prior to blocking with 10% FCS for 30 minutes at room temperature. The primary antibody was diluted 1/1000 and incubated with the sample for 2 hours. The secondary antibody was an Alexa Fluor® 488-conjugated Goat antibody, diluted 1/1000.See Abreview
