The blot shown above was stripped and reprobed with pan-Aurora specific polyclonal antibody (available as ab12875) to verify equal loading of Aurora A in the extracts. Proteins were visualized using a donkey anti-rabbit secondary conjugated to HRP (available as ab16284)and a chemiluminescence detection system.
Immunofluorescence analysis of P. mammillata embryos during mitotic cycle, staining Aurora A (phospho T288) with ab18318.Embryos were fixed in methanol/1%formaldehyde (-20°C) overnight, permeabilized in 0.25% Triton X-100 for 15 minutes at room temperature and then incubated in primary antibody (1/200) in PBS/1%BSA overnight at 4°C. A FITC-conjugated anti-rabbit IgG (1/50) was used as the secondary antibody.
Prometaphase HeLa cells were permaeabiliased in 0.2% Triton-X100 and fixed for 10 minutes at room temperature in 4% formaldehyde. The cells were washed 3 times in PBS and incubated with ab18318 (1/1000) for 1 hour at room temperature.
