Immunofluorescent analysis of U2OS cells showing labeling of CDT1 with ab183478 at 1/2000 dilution. Anti-Cyclin B labeling is seen in red and DAPI counterstain in blue. Lower image is merged.The cells were fixed in PBS containing 3% paraformaldehyde for 10 min, permeabilized for 2 min with cold methanol and washed 2 or 3 times with PBS. The cells were blocked in 2% BSA-PBS-Tween 20 0.2% (BSA-PBS-T) for 1h @ room temperature. The cells were incubated with ab183478 at 1/2000 in BSA-PBS-T overnight @ 4C. They were washed 3-4X with PBS-T (PBS-Tween 20 0.2%), incubated with secondary in BST-PBS-T 1 h @ room temperature, washed 3-4X with PBS-T and then washed 2X with PBS only.
