Specificity of MIP2 on RPMI cell culture media and PBMC conditioned supernatants. Human PBMCs were cultured in RPMI supplemented with 10% fetal calf serum, 2 mM L-glutamine, 100 U/mL penicillin, and 100 µg/mL streptomycin. Cells were cultured for 2 days at 37⁰C in the present or absence of PHA. The concentrations of MIP2 were interpolated from a standard curve in base RPMI. The mean MIP2 concentration was determined to be undetectable in basal media, 468.3 pg/mL in unstimulated PMBC supernatants and 671.6 pg/mL in stimulated PBMC supernatants.
The levels of MIP2 in serum samples were tested from ten individual healthy donors. Levels were interpolated from the standard curve in Sample Diluent 50BP. The mean level of MIP2 is 459.2 pg/ml with a range of 22.2 pg/mL to 1773 pg/mL and a standard deviation of 512.1 pg/mL.
