Description Vincristine is a vinca alkaloid isolated from the plant Catharanthus roseus that acts as a mitotic inhibitor blocking microtubule assembly in vitro (1,2). Research studies demonstrated that vincristine binds to and inhibits the addition of tubulin dimers to the assembly end of steady-state microtubules in a dose-dependent manner (Ki = ~85 nM) (1). Additional research suggests that high concentrations of vincristine depolymerize microtubules (2). Disruption of mitotic spindle function has been shown to cause mitotic arrest, inhibit cell proliferation, and induce apoptosis in numerous cell types (1-6).
