| Bioactivity | Enzyme activity is assayed by an AmplexRed/Peroxidase coupling method using 1,5-diaminopentane as a substrate. Purified LOXL-4 (0.25-1 ng/ul) is incubated with 3 mM 1,5-diaminopentane, 0.4 mM AmplexRed and 0.2 unit/ul peroxidase in 100 μl of the assay buffer (1.2M Urea, 50 mM borate pH=8.8) for 15 min at 30 oC. Fluorescence intensity is measured every 7 minute at 530ex/590em. |
