SEE ALSO...

NIPA Antibody (B-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects NIPA protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-NIPA antibody (B-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Nuclear Interaction Partner of ALK (NIPA) protein plays a pivotal role in regulating cyclin B1, which is essential for proper cell cycle progression. NIPA is primarily located in cytoplasm during interphase, where phosphorylation occurs during G2 and M phases, leading to cyclin B1 accumulation in the nucleus just before mitosis. This phosphorylation prevents premature mitotic entry by allowing cyclin B1 to accumulate until appropriate signaling occurs. When NIPA dephosphorylates, interaction with the SCF complex facilitates ubiquitination and subsequent degradation of cyclin B1, thus maintaining genomic stability. Dysregulation of NIPA can lead to abnormal cyclin B1 accumulation, resulting in premature mitotic entry and loss of checkpoint control, which are significant factors in cancer development. Additionally, phosphorylated NIPA may also play a role in apoptotic signaling pathways, highlighting NIPA′s importance in both cell cycle regulation and programmed cell death.

What is the recommended fixation method for immunofluorescence staining with NIPA (B-10): sc-365058 monoclonal antibody?

Asked by: Dr Ninau Qelp
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
