ATF-5 Antibody (E-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects ATF-5 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). ATF-5 (E-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ATF-5 plays a crucial role in cellular stress responses and is involved in the regulation of gene expression during conditions such as hypoxia and nutrient deprivation. ATF-5′s ability to modulate transcription in response to stress is vital for cell survival and adaptation, making ATF-5 an important target for research in cancer and neurodegenerative diseases. ATF-5 localizes in both the cytoplasm and nucleus, where ATF-5 functions as a dimer to bind DNA, and interacts with other proteins such as CCND3 and PTP4A1, which are involved in cell cycle regulation and signal transduction pathways. The ATF/CREB transcription factor family, to which ATF-5 belongs, is characterized by a conserved C-terminal leucine zipper that facilitates dimerization and DNA binding, while the N-terminal domains exhibit significant variability, allowing for diverse regulatory functions.

Hi, I am interested in purchasing this antibody for an immunofluorescence. However, I feel like there isn't enough information in the caption provided to test it out. Can you direct me to the protocol for this staining? Megan

I am using ATF-5 (E-10): sc-377168 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Cweed
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 4 out of 5 by from Good work!!This antibody gave a strong and clean signal when we used it at a 1:100 diluition with 80μg of protein samples.

Rated 4 out of 5 by from Specific for ATF5 with a close non-specific bandWorks nicely for western blotting of HeLa lysates at a 1:500 dilution at 4 degrees overnight, producing a clean, specific band at approximately 38kDa. However, there is a strong non-specific band that runs very close to the ATF5 band at just above 40kDa.
