Bcl-6 Antibody (H-12) is a mouse monoclonal IgG1 kappa light chain antibody designed to detect Bcl-6 protein, also known as BCL6 transcription repressor or B-cell lymphoma 6 protein, across mouse, rat, human, equine, canine, bovine, and porcine species, utilizing applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Bcl-6 monoclonal antibody (H-12) specifically targets an epitope mapping to amino acids 679-706 at the C-terminus of human Bcl-6 protein, ensuring precise and reliable binding. Bcl-6 (H-12) antibody recognizes a pivotal transcriptional repressor involved in regulating gene expression by binding to Stat recognition-like DNA elements, playing an essential role in germinal center development, B cell differentiation, and T helper cell differentiation into Th1 and Th2 subsets, which are critical for adaptive immune response. Structurally, Bcl-6 contains a BTB/POZ domain responsible for protein-protein interactions and multiple zinc finger motifs that facilitate specific DNA binding, enabling interactions with various co-repressors such as SMRT and BCOR, which are crucial for transcriptional repression. These interactions are vital for modulating processes like NFκB signaling, where Bcl-6 negatively regulates NFκB expression, thereby inhibiting NFκB-mediated cellular functions important for maintaining cellular homeostasis and preventing excessive inflammatory responses. Post-translational modifications of Bcl-6, including acetylation, phosphorylation, and ubiquitination, are critical for regulating stability, localization, and transcriptional activity, with HDAC- and SIR2-dependent acetylation specifically downregulating Bcl-6 activity by inhibiting histone deacetylase complex recruitment. Dysregulation of Bcl-6 through aberrant post-translational modifications is frequently observed in non-Hodgkin′s B cell lymphomas, highlighting significance in oncogenesis and making Bcl-6 a valuable target for therapeutic intervention. Anti-Bcl-6 antibody (H-12) is available in both non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates.
