PKAα cat Antibody (A-2) is a mouse monoclonal IgG2a kappa light chain antibody engineered to target the protein kinase A (PKA) alpha catalytic subunit (PRKACA) with exceptional precision. PKAα cat monoclonal antibody (A-2) recognizes an epitope mapped to amino acids 322-351 at the C-terminus of the human PKAα catalytic subunit, ensuring robust detection in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications across multiple species, including mouse, rat, and human, as well as additional species such as equine, bovine, and porcine, though may exhibit cross-reactivity with the β and γ subunits of PKA. PKA plays a pivotal role in cellular signaling by mediating responses to the second messenger cyclic AMP (cAMP), which is essential for regulating a wide array of physiological processes including cell proliferation, ion transport, metabolism, and gene transcription. Upon binding of cAMP, the regulatory subunits of the tetrameric PKA holoenzyme dissociate, releasing the active catalytic subunits that phosphorylate target proteins such as the transcription factor CREB on serine 133. This phosphorylation event is critical for the initiation of gene transcription, linking extracellular signals to the regulation of gene expression and ultimately influencing cellular function and behavior. Anti-PKAα cat antibody (A-2) is available in both non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatile options to accommodate diverse experimental requirements and facilitating advanced studies into the mechanisms of PKA-mediated cellular processes.

The exact peptide sequence used to develop this monoclonal antibody is considered proprietary. That is why there was inconsistent information on our website and product data sheet. For consistency, we will update our product information with the following amino acid range: peptide mapping between amino acids 322-351 at the C-terminus. The exact epitope recognized by this mouse monoclonal antibody has not yet been determined.

Hello, My lab used to use this polyclonal antibody and since it is being stopped we tested your monoclonal antibody. However, the monoclonal had nonspecific binding, do you know why that is?

The antibody also generates a clean signal for Western blotting (enhanced chemiluminescence). In addition, sc-28315 is suitable for immunoprecipitation.
