IL-6 Antibody (B-F6) is a mouse monoclonal IgG1 antibody that detects interleukin-6 in human samples through western blotting (WB) and immunoprecipitation (IP) applications. IL-6 is a multifunctional protein composed of 212 amino acids, playing a pivotal role in host defense, immune response, and hematopoiesis. IL-6 is constitutively expressed by epidermal Langerhans cells, with expression further induced in stimulated keratinocytes, highlighting importance in the skin′s immune response. Notably, IL-6 acts as an endogenous pyrogen alongside IL-1β and tumor necrosis factor-alpha (TNFα), regulating fever response during bacterial infections. The IL-6 receptor is a trimeric complex that includes an IL-6-specific alpha chain and a homodimer of the gp130 glycoprotein, which is common to several other receptors, including those for IL-11, ciliary neurotrophic factor (CNTF), oncostatin M (OSM), and leukemia inhibitory factor (LIF). Upon IL-6 binding, the receptor complex undergoes homodimerization of gp130, leading to activation of Janus kinases JAK1 and JAK2. These kinases subsequently phosphorylate signal transducer and activator of transcription 3 (Stat3), facilitating nuclear translocation and transcription of Stat3-responsive genes, which are crucial for various cellular responses. Additionally, IL-6 has been implicated in activating the Ras/MAP kinase pathway, further influencing transcription of NFIL6, thereby underscoring significance in both inflammatory responses and cellular signaling pathways.
