ATF3 (activating transcription factor 3) belongs to ATF/CREB family of transcription factors and it binds Cre 5'-GTGACGT[AC][AG]-3' a sequence which is present in several cellular as well as viral promoters. ATF3 exerts its repressive effects on transcription from promoters with ATF sites by stabilizing the binding of inhibitory cofactors of target promoters. Expressed by various cell types with nuclear localization, ATF3 expression gets upregulated by a variety of extracellular stimuli such as cytokines, chemokines, growth factors, hormones, hypoxia, DNA damage, ER stress and starvation. Originally considered a stress-response gene, ATF3 now has emerged as a factor implicated in general adaptive responses such as environmental, emotional and nutritional alterations. Its disorders have been associated with inflammatory and immune diseases as well as cancer, and unlike other ATF/CREB family members, ATF3 is emerging as a factor involved in protection against malignant cells and invading pathogens.

We have a customer who is interested in the following antibodies: NB600-904 c-Fos ; NBP1-02935 ATF3. She is not sure which ones would be suitable for her studies. Her message is as follows: I have a new list of antibodies I would like you to check for me. We need them for paraformaldehyde perfused mice, the DRGs (dorsal root ganglia) cut by frozen section. So not paraffin processed. Would Novus be able to comment on the suitability of these antibodies for our tissues. I would also be grateful if they could make other suggestions if they have other more suitable antibodies or comment on which of the two Il1b antibodies would be the better. I am hoping you are able to help us with our customer. ..show answer..
While NBP1-02935 has not been tested on frozen sections, none of our other ATF3 antibodies have been tested either. NBP1-02935 works fine in IHC on paraffin embedded tissues, so should be suitable for use on frozen sections as well, although we can not guarantee that. Generally an antibody that is suitable for paraffin-embedded sections should work on frozen sections as well, however, we have seen evidence that this is not always true. This is why we make a distinction. The customer is more than welcome to try an IHC-P guaranteed antibody on frozen sections if they would like since it will most likely work, however, we can not guarantee it for this use. NB110-61652 is sheep polyclonal, not NB600-904 and once again, NB600-904 isn't guaranteed for use in frozen sections but the customer can certainly give it a try. I recommended NB110-75039 since it does have a frozen tissue guarantee. NB110-75039 and NB110-61652 are both guaranteed to detect c-Fos on frozen sections. I would recommend NB110-75039 since we do have images for this product and it is a rabbit polyclonal, so it is much more likely the customer would have an anti-rabbit secondary available rather than an anti-sheep secondary.
