SEE ALSO...

Calgranulin A/B Antibody (27E10) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Calgranulin A/B protein of human origin by immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Anti-Calgranulin A/B antibody (27E10) is available in both non-conjugated and various conjugated forms, including phycoerythrin (PE) and fluorescein isothiocyanate (FITC), providing versatility for different experimental needs. Calgranulin A/B, also known as S100A8/S100A9, plays a crucial role in the immune response, particularly in the regulation of inflammation and the recruitment of immune cells to sites of injury or infection. S100A8/S100A9 is primarily expressed in myeloid cells, such as macrophages and neutrophils, and is involved in the formation of heterodimers that can modulate various cellular processes, including cell proliferation and apoptosis. S100A8/S100A9 presence in inflammatory conditions makes this protein a significant biomarker for diseases such as cystic fibrosis and rheumatoid arthritis, highlighting S100A8/S100A9′s importance in both diagnostic and therapeutic contexts. Calgranulin A/B monoclonal antibody (27E10) enables researchers to gain valuable insights into the mechanisms of inflammation and immune regulation, furthering our understanding of various pathologies.

Can Calgranulin A/B (27E10): sc-33714 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: DefinitelyNotMatt
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Calgranulin A/B (27E10): sc-33714 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

How did you determine that the antibody is binding Calgranulin A/B? Do you know the epitope? Does it bind A or B ? i.e. do you know if it is binding the S100A8, S100A9 or S100A12 subunits ? Thanks!

Asked by: Mark R
Thank you for your inquiry. Our Calgranulin A/B antibody sc-33714 was raised against human monocytes, but the epitope has not been mapped. One publication, PubMedID 1529531, describes an intracellular epitope. This is a commercially available clone that has been determined to be Calgranulin A/B reactive. We do not have information specific to the reactivity with S100A8, S100A9 or S100A12 subunits.
