MAD1 Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects MAD1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MAD1 (F-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MAD1 plays a critical role in mitotic spindle assembly checkpoint, ensuring cell cycle progression halts until all chromosomes properly align at metaphase plate. This function maintains genomic stability, as improper spindle assembly can lead to aneuploidy and contribute to cancer development. MAD1 interacts with MAD2 to form a tetrameric core complex, essential for checkpoint function. MAD1 primarily localizes in nucleus but redistributes to centrosome and spindle midzone during mitosis, highlighting dynamic role in cell division. Phosphorylation regulates MAD1 activity, particularly in response to spindle damage, and MAD1 gene defects are implicated in various cancer types, underscoring importance in both normal cellular function and disease progression.
