ab11820 staining HLA E in Human endothelial cell line by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 5% BSA for 2 hours at 4°C. Samples were incubated with primary antibody (1/100 in 5% BSA in PBS with 0.1% Triton X-100) for 1 hour at 4°C. An undiluted FITC-conjugated Goat anti-mouse polyclonal was used as the secondary antibody.See Abreview
Cells were treated with 500 Units of human IFN-gamma on 1 million ECV304 cells after 36 hours cells were harvested (PVG 0.1% trypsin), fixed with 2% paraformaldehyde and surface staining of HLA E was performed using ab11820 at a 1/100 dilution. The secondary used was a goat anti-mouse-FITC, at a 1/100 dilution.Red line indicates the untreated expression level and black indicate the induced surface expression of HLA E. The lysates were immunoblotted with antibodies against HLA-G, HLA-E (ab11820), or beta-actin as a loading control. The figure shows that expression of HLA-G is slightly reduced by US10.
