ab72311 staining VGLUT1 in synaptosomes obtained from human nerve cells by Flow Cytometery. Samples were prepared by homogenization and centrifugation in sucrose. Cells were fixed in formaldehyde and permeabilized in 0.2% Tween. The primary antibody was diluted to 0.5 µg/ml (PBS + Tween) and incubated with sample for 30 minutes at 25°C. Immunofluorescence detection of VGLUT1 using ab72311 on rat primary hippocampal neurons. Fixative: Paraformaldehyde. Permeabilization: Triton 0.3% in 0.1X PBS. Blocking step: 10% serum for 20 mins @ 24°C. Primary Antibody Concentration: 1 µg/ml (for 1 hour @ 24°C) in 10% Donkeys serum, Triton 0.3%. Secondary Antibody Name: Alexa Fluor® 488 (1/1000). ICC/IF image of ab18258 stained primary hippocampal rat neurons. The cells were 4% PFA fixed (10 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab18258, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 and VGLUT2 (ab79157)(red). ICC/IF image of ab72311 stained PC12 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab72311, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
