Semaphorin 4C in Human Brain. Semaphorin 4C was detected in immersion fixed paraffin-embedded sections of human brain (cortex) using Sheep Anti-Human Semaphorin 4C Antigen Affinity-purified Polyclonal Antibody at 15 ug/ml overnight at 4°C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic. Tissue was stained using Anti-Sheep HRP-DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to neuronal cell bodies.
Detection of Human Semaphorin 4C by Western Blot. Western blot shows lysates of SW13 human adrenal cortex adenocarcinoma cell line. PVDF membrane was probed with 0.5 ug/ml of Sheep Anti-Human Semaphorin 4C Antigen Affinity-purified Polyclonal Antibody followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody. A specific band was detected for Semaphorin 4C at approximately 110 kD (as indicated). This experiment was conducted under reducing conditions.
