Immunofluorescence analysis of Human corneal endothelial cells, staining Cyclin E with ab7959.Cells were fixed with methanol, permeabilized with n.a and blocked with serum for 30 minutes at 37°C. Samples were incubated with primary antibody (1:200 in 2% BSA + 2% goat serum) for 1 hour at 37°C. An undiluted AlexaFluor®555-conjugated goat anti-rabbit polyclonal IgG (ab150078) was used as the secondary antibody.See Abreview
ab7959 staining Cyclin E in murine C2C12 cells by Immunocytochemistry/ Immunofluorescence. The cells were fixed in paraformaldehyde, permeabilised in 0.3% Triton X-100 and then blocked using 2% serum for 1 hour at 25°C. Samples were then incubated with primary antibody at 1/500 dilution for 16 hours at 4°C. The secondary antibody used was a goat anti-rabbit IgG (H+L) (ab150078) conjugated to Alexa Fluor® 594 (red) used at a 1/1000 dilution.See Abreview
Ab7959 staining Human normal placenta. Staining is localized to the nucleus.Left panel: with primary antibody at 1 ug/ml. Right panel: isotype control.Sections were stained at room temperature. Sections were rehydrated and antigen retrieved. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 minutes. They were then blocked for 10 minutes (containing casein 0.25% in PBS), then incubated with primary antibody for 20 minutes, and detected 30 minutes. Colorimetric detection was completed with diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.
