Function Plays a role in the regulation of the translocation of GLUT4 to the cell surface in adipocytes and skeletal muscle cells in response to insulin, probably by regulating RAB31 activity, and thereby contributes to the regulation of insulin-dependent glucose uptake (By similarity). Low affinity receptor which can bind to NGF, BDNF, NT-3, and NT-4. Can mediate cell survival as well as cell death of neural cells. ICC/IF image of ab87472 stained PC12 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA, 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab87472 at 1/100 dilution overnight at +4°C. The secondary antibody (green) was DyLight® 488 donkey anti- goat (ab96931) IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Immunocytochemistry/ Immunofluorescence of HCEM cells labelling p75 NGF Receptor (green) with ab25958 at 1/500. Cells were cultured until low confluence on 35mm glass base dishes and washed with PBS. Cells were fixed with 4% paraformaldehyde in PBS and blocked by incubation with TBS containing 0.2% casein and 0.1% Triton X-100 for 20 minutes. Cells were incubated with primary antibody (1/500) overnight at 4°C. An Alexa Fluor® 488 anti-rabbit IgG (1/200) was used as the secondary antibody. Beta actin was stained with Alexa Fluor® 594 phalloidin (red).

Ab46173 staining p75 NGF Receptor in rat spinal cord by immunohistochemistry using frozen tissue. Overlay histogram showing SH-SH5Y cells stained with ab93934 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab93934, 1/100 dilution) for 30 min at 22°C. The secondary antibody used was Alexa Fluor® 488 goat anti-mouse IgG (H&L) (ab150113) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 1μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in SH-SY5Y cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
