Function Thought to form a receptor-activated non-selective calcium permeant cation channel. Probably is operated by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases or G-protein coupled receptors. Activated by diacylglycerol (DAG) in a membrane-delimited fashion, independently of protein kinase C. Seems not to be activated by intracellular calcium store depletion.

Tissue Specificity Expressed primarily in placenta, lung, spleen, ovary and small intestine. Expressed in podocytes and is a component of the glomerular slit diaphragm. {ECO:0000269|PubMed:15924139}.

ab12249 staining TRPC6 in mouse Olfactory tissue section by Immunohistochemistry (PFA perfusion fixed frozen sections). Mice were anesthetized with 20% chloral hydrate (2 mg/g body weight), perfused transcardially with 0.9% saline followed by 4% paraformaldehyde in 0.1 M phosphate buffer (PB). The olfactory organs were dissected and postfixed in the same fixative for 15 min to overnight. Cryosections were rinsed in 0.1 M phosphate buffered saline (PBS), blocked in blocking solution containing 1% BSA, 3% normal donkey serum, and 0.3% Triton X-100 in PBS for 2 hours, and then incubated in the primary antisera overnight to 2 days. An Alexa Fluor® 568 conjugated anti rabbit IgG was used for 2 hours as secondary. ab12249 stains red in image shown.
